gstic acid hematoxylin solution
Hematoxylin-1g
Phosphotungstic acid-20g
Distilled water-1000ml
Procedure
①탈수와 탈파라핀화
②5% acetic acid가 포함된 Zenker\'s solution을 3시간 반동안 56~60도 오븐에서 매염처리한다.
③오븐에서 꺼내고 30분간 차갑게 처리한다.
④iodine으로 mercuric chloride crystal을 제거하고, sodium thiosulfate로 투명화한다.
⑤흐르는 물에서 10분간 수세한다.
⑥56~60도 오븐에서 90분간 PATH용액에 염색한다.
⑦꺼낸뒤 냉각시킨다.
⑧95% alcohol로 빠르게 탈수, 100% alcohol로 탈수, 자일렌으로 투명화를 각각 2번 시행한다.
⑨permount나 histoclad로 mounting한다.
