희석된 sample 소량을 빈 plate상에 먼저 떨어뜨린 후, 액화된 상태의 LB agar를 부어 굳힌다.

LB agar plate, loop, alcohol lamp, Incubator, test tube, pipette man, O/N 배양된 E. coli
* LB배지의 조성
Bacto-tryptone : 1 %
Yeast extracts : 0.5 %
NaCl : 1 %
IV. Procedure - Single colony isolation
I) Streak plate methods
II) Pour plate methods
① LB culture에 E. coli를 O/N 배양한다.
② Dilution method를 사용하여 sample을 희석한다.
③ 10-1, 10-2 10-3등으로 serial dilution 한 것을 빈 plate상에 떨어뜨린다.
④ 액화된 상태의 LB agar를 plate에 적당량 (약 25 ml)붓고, 천천히 흔든 후 굳힌다.
⑤ 37℃ Incubation에서 배양한 후 관찰한다.
