Phenol-Sulfuric Acid Method for Total Carbohydrates
Introduction:
The total carbohydrate contents of liquid food and food extracts can be determined. Simple sugars, oligosaccharides, polysaccharides, and their derivatives give a stable orange color when reacted with phenol and concentrated sulfuric acid.
Simple sugars + Phenol + Sulfuric Acid ------- > Orange-yellow color
The intensity of the orange color is proportional to the amount of total carbohydrates present and its absorbance can be measured at 492 nm. The level of total carbohydrates in the test solution can be determined by reference to the standard curves prepared.
Before coming to the lab, review the principles of standard curves.
